Project Goal
The goal of this project is to develop an algorithm that will effectively and predictably increase the expression level of a given native sequence of a target protein, solely by altering codons of each amino acid. The deliverable will consist of a repeatable method of increasing the protein expression, therefore allowing the optimization and cost reduction of the protein of interest. After validation of our model the group hopes to compare our model to other available codon optimizer models, such as IDT.
Project Objectives
The group will attempt to accomplish the project goal by following the milestone objectives defined as follows:
-Compile a working library that contains a large sample of protein sequences and their respective yields. Identify quantitative parameters to be performed on the primary sequences of each protein in the library. Normalize these parameters.
- Create MatLab scripts to calculate each of these parameters. Develop an algorithm within MatLab to run these scripts for every data set within the library, and group this information in an array with which to analyze.
-Perform statistical regression of the parameters against their correlation with improving protein expression. Identify the key parameters most influential in increasing yield.
-Develop model to optimize the key parameters identified, thus optimizing the protein production.
